Identity and purity testing for thymosin alpha 1 relies mainly on reversed-phase high-performance liquid chromatography and mass spectrometry. Chromatography separates the parent peptide from truncated or modified variants, while mass spectrometry confirms the expected molecular mass. Amino acid analysis and peptide mapping provide additional sequence confirmation. Counterion content, water content, and residual solvents are measured separately as part of specification testing. No single method captures every attribute, so laboratories combine several techniques.
The peptide lacks cysteine, methionine, and tryptophan, so disulfide scrambling and sulfur oxidation are not major degradation routes. Instead, aspartate residues can undergo isomerization or cyclization to succinimide intermediates, generating isoaspartate variants. Hydrolysis of peptide bonds also occurs slowly in solution. These changes may reduce biological activity even when the main peak remains detectable. Stability studies therefore track both potency and the appearance of related substances.
Identity and purity are assessed with a small set of standard peptide methods. Reversed-phase high-performance liquid chromatography separates the main peak from truncated or oxidized species, and its area percentage is the usual purity measure. Mass spectrometry confirms the observed molecular mass against the expected value, while amino acid analysis or peptide mapping checks composition and sequence. Specifications for research-grade material are often stated as 95 percent or higher, though the exact limit depends on the supplier and the intended use.
Laboratory supplies of the peptide usually arrive as a lyophilized powder in sealed vials. The powder is hygroscopic, so a vial should be allowed to reach room temperature before it is opened to prevent condensation on the contents. Weighing and transfer are best performed in a low-humidity environment with clean tools. Once dissolved, the solution should be mixed gently rather than vortexed, because foaming and shear can reduce recovery of the peptide.
Storage recommendations center on low temperature, dryness, and protection from repeated freezing and thawing. The intact powder is commonly held at 20 degrees below zero Celsius or colder, while a working solution is divided into single-use aliquots to limit freeze-thaw cycles. Buffered saline or phosphate-buffered saline at neutral pH is frequently used as a diluent. Light sensitivity is not well documented, yet amber vials or foil wrapping are common practice for long-term storage of peptide stocks.

Practical handling focuses on limiting adsorption and contamination. The peptide dissolves readily in water, and dilute solutions tend to adhere to plastic and glass surfaces, so an inert carrier protein or a defined buffer can reduce losses in laboratory work. Workers also record the counter-ion form, since an acetate or trifluoroacetate salt changes the mass balance of the weighed powder. Documentation of lot number, purity value, and storage history supports reproducibility when results from different laboratories are compared.
Lyophilized material is generally held at reduced temperature to slow degradation, and storage at minus twenty degrees Celsius or lower is common practice for long-term retention. Short-term working portions are often kept between two and eight degrees Celsius. Once dissolved, the peptide is less stable than the dry powder, and repeated freeze-thaw cycles are associated with loss of material and with aggregate formation. Vials are usually allowed to reach room temperature before opening so that condensation does not introduce moisture, and solutions are protected from light where practical.
=== Skin === The most common side effects are mucocutaneous: dry lips, skin, and nose. Other common mucocutaneous side effects are inflammation and chapping of the lips (cheilitis), redness of the skin (erythema), rashes, peeling, eczema (dermatitis), itching (pruritus) and nose bleeds (epistaxis). Absence of dryness of the lips is considered an indication of non-compliance with treatment (not taking the drug as advised), as it occurs in almost all people who take it. Regular use of lip balm and moisturizer is recommended throughout treatment to reduce these problems. The dose may need to be decreased to reduce the severity of these side effects. The skin becomes more fragile—especially to frictional forces—and may not heal as quickly as normal. Wound healing is delayed. For this reason, elective surgery, waxing of hair, tattooing, tattoo removal, piercings, dermabrasion, exfoliation, etc., are not recommended during treatment. Treatment of acne scars is generally deferred until 12 months after completion of a course of isotretinoin.
In molecular biology, a termination factor is a protein that mediates the termination of RNA transcription by recognizing a transcription terminator and causing the release of the newly made mRNA. This is part of the process that regulates the transcription of RNA to preserve gene expression integrity and are present in both eukaryotes and prokaryotes, although the process in bacteria is more widely understood. The most extensively studied and detailed transcriptional termination factor is the Rho (ρ) protein of E. coli.
=== Counting methods === In order to quantify the number of substitutions, one may reconstruct the ancestral sequence and record the inferred changes at sites (straight counting – likely to provide an underestimate); fitting the substitution rates at sites into predetermined categories (Bayesian approach; poor for small data sets); and generating an individual substitution rate for each codon (computationally expensive). Given enough data, all three of these approaches will tend to the same result.
Even the shell structure in the nucleus and electron cloud of oganesson is strongly impacted by relativistic effects: the valence and core electron subshells in oganesson are expected to be "smeared out" in a homogeneous Fermi gas of electrons, unlike those of the "less relativistic" radon and xenon (although there is some incipient delocalisation in radon), due to the very strong spin–orbit splitting of the 7p orbital in oganesson. A similar effect for nucleons, particularly neutrons, is incipient in the closed-neutron-shell nucleus 302Og and is strongly in force at the hypothetical superheavy closed-shell nucleus 472164, with 164 protons and 308 neutrons. Studies have also predicted that due to increasing electrostatic forces, oganesson may have a semibubble structure in proton density, having few protons at the center of its nucleus. Moreover, spin–orbit effects may cause bulk oganesson to be a semiconductor, with a band gap of 1.5±0.6 eV predicted. All the lighter noble gases are insulators instead: for example, the band gap of bulk radon is expected to be 7.1±0.5 eV.
Conversely, other researchers have argued that the risks of progestogens in transgender women are likely minimal, and that in light of potential albeit hypothetical benefits, should be used if desired. In general, some transgender women respond favorably to the effects of progestogens, while others respond negatively. A 2025 study found that 72% of progesterone users were satisfied with progesterone use, and 73.5% would recommend progesterone to others. Progesterone is most commonly taken orally. However, oral progesterone has very low bioavailability, and produces relatively weak progestogenic effects even at high doses. In accordance, and in contrast to progestins, oral progesterone has no antigonadotropic effects in males even at high doses. Progesterone can also be taken by various parenteral (non-oral) routes, including sublingually, rectally, and by intramuscular or subcutaneous injection. These routes do not have the bioavailability and efficacy issues of oral progesterone, and accordingly, can produce considerable antigonadotropic and other progestogenic effects. Transdermal progesterone is poorly effective, owing to absorption issues. Progestins are usually taken orally. In contrast to progesterone, most progestins have high oral bioavailability, and can produce full progestogenic effects with oral administration. Some progestins, such as medroxyprogesterone acetate and hydroxyprogesterone caproate, are or can be used by intramuscular or subcutaneous injection instead. Almost all progestins, with the exception of dydrogesterone, have antigonadotropic effects.

=== Series of coups d'état === On 10 February 1988, Rocky Malebane-Metsing of the People's Progressive Party (PPP) became the president of Bophuthatswana for one day when he took over the government through a military coup. He accused Mangope of corruption and charged that the recent election had been rigged in the government's favour. A statement by the defence force said "serious and disturbing matters of great concern" had emerged, citing Mangope's close association with a multimillionaire Israeli Soviet émigré Shabtai Kalmanovich. Subsequently, the South African Defence Force invaded Bophuthatswana and Mangope was reinstated and continued his term unabated. P. W. Botha, State President of South Africa at the time, justified the reinstatement by saying that "[t]he South African Government is opposed in principle to the obtaining or maintaining of power by violence." In 1990, a second coup attempt took place in which an estimated 50,000 protesters demanded the President's resignation over his handling of the economy. The New York Times reported that seven people had been killed and 450 wounded "after police officers in armoured cars fired their rifles into the crowds and used tear gas and rubber bullets". After Mangope had asked for help from the South African government, he declared a state of emergency and cut telephone links to the territory "for political reasons", claiming that "normal laws had become inadequate". Human Rights Watch put the number of protesters at 150,000.

The lyophilized powder is kept refrigerated at 2 to 8 degrees Celsius and protected from light. Reconstituted solutions should be used promptly. Freezing and thawing repeatedly is avoided.
Reversed-phase HPLC is the primary tool for purity, paired with mass spectrometry for identity. Amino acid analysis and peptide mapping add sequence confirmation. Several techniques are combined because no single test covers every attribute.
It lacks sulfur-containing residues, so oxidation is limited. Aspartate isomerization and slow hydrolysis are the main concerns. Solution stability is shorter than that of the lyophilized powder.
Aliquots are typically frozen well below zero Celsius and thawed only once, since repeated cycles promote aggregation and loss. Dilution into a neutral buffer limits degradation during short working periods, and prolonged storage at room temperature is avoided.
